双波长吸收光谱法测定地奥司明

摘    要:建立了测定药物中地奥司明含量的双波长吸收光谱法.以碱性艳蓝B作探针,在pH 5.48 Tris-HCl缓冲溶液中,用双波长吸收光谱法定量检测地奥司明,测定波长为(292+442) nm.在弱酸性溶液中,碱性艳蓝B与地奥司明反应生成蓝色二元离子缔合物.在250-530 nm范围内,谱线上有2个强度较大的正吸收峰,分别位于292和442 nm,表观摩尔吸光系数(κ)分别为3.40×104 L/(mol·cm)和8.06×10 4L/(mol·cm).当用双波长法测定时,表观摩尔吸光系数(κ)为1.15×105 L/(mol·cm),加标回收率为98.5%-103%,相对标准偏差(RSD)(n=5)为2.2%-2.6%.线性范围均为0.3-15.2 mg/L.实验表明,用双波长法测定地奥司明含量,不仅有高的灵敏度,且方法简便、快速,适于市售地奥司明片剂中地奥司明含量的测定.

Abstract:To establish a new dual-wavelength absorption spectrometry for the determination of diosmin in drugs samples,quantitative detection of diosmin was performed by dual-wavelength absorption spectroscopy in pH 5.48 Tris-HCl buffer solution using basic brilliant blue B as probe with a detection wavelength of(292+442)nm.In a weak acidic solution,basic brilliant blue B reacted with diosmin to form a blue binary ion associate complexes.In the range of 250 to 530 nm,the new material has two large positive absorption peaks located at 292 and 442 nm and their apparent molar absorbance coefficient (κ)were 3.40×104 L/(mol·cm)and 8.06×104 L/(mol·cm),respectively.When measured by using the dual-wavelength method,the apparent molar absorbance coefficient(κ)can reach 1.15×105 L/(mol·cm),the spiked recovery was 98.5%~103% and the relative standard deviation(RSD)(n=5)was 2.2%~2.6%.Their linear range were from 0.3 to 15.2 mg/L.Experiments showed that the dual-wavelength method used for the determination of diosmin has not only high sensitivity,but also a simple and rapid operation so that it is suitable for the determination of diosmin in commercially available diosmin tablets.
