大粒车前子多糖对树突状细胞分泌不同类型细胞因子的影响

初步探讨大粒车前子精制多糖（PL-PS）对树突状细胞分泌不同类型细胞因子的影响。通过体外细胞因子联合诱导培养小鼠骨髓细胞获得树突状细胞（DCs）。经免疫磁珠分选法纯化获取CD11c＋树突状细胞，经不同浓度PL—PS刺激24h后，采用酶联免疫吸附法（ELISA）检测培养上清液中细胞因子IL-12pTO、IL-10及IL-1β含量。结果表明：在质量浓度25—100μg／mL范围内，PL-PS均以剂量依赖的方式上调DCs分泌IL-12pTo和IL-1B水平、下调IL-10的分泌水平。可因此得出推论：PL—PS能够通过调控DCs分泌Th1、Th2型细胞因子及趋化因子水平，诱导Th1型细胞免疫应答。

Effects of Polysaccharides Isolated from the Seeds of Plantago Asiatica L. on Cytokine Production of Dendritic Cells

To investigate the effects of polysaccharides isolated from the seeds of Plantago asiatica L. （PL-PS） on the secretion activity of bone marrow-derived dendritic cells （DCs）. Bone marrow-derived dendritic cells were induced by 5-day-cuhure in RPMI-1640 medium supplemented with 10% heat-inactivated fetal calf serum, recombi- nant mouse granulocyte-macrophage colony-stimulating factor and interleukin-4 ,and then CD1 1c ~ cells isolated by MACS were cultured for 24 h in the presence of PL-PS at varying concentrations. The levels of IL-12p70, IL-10 and IL-1 β in culture supematant of DCs were detected by ELISA. Results showed that PL-PS up-regulated the secretion of IL-12p70 and IL-1β ,down-regulated the secretion of IL-10 in a dose-dependent manner at 25 -100μg/mL. The effects of PL-PS on the cytokine secretion of DCs suggested that PL-PS may play a role in inducing cellullar immu- nologic response.
