摘    要:初步探讨大粒车前子精制多糖(PL-PS)对树突状细胞分泌不同类型细胞因子的影响。通过体外细胞因子联合诱导培养小鼠骨髓细胞获得树突状细胞(DCs)。经免疫磁珠分选法纯化获取CD11c+树突状细胞,经不同浓度PL—PS刺激24h后,采用酶联免疫吸附法(ELISA)检测培养上清液中细胞因子IL-12pTO、IL-10及IL-1β含量。结果表明:在质量浓度25—100μg/mL范围内,PL-PS均以剂量依赖的方式上调DCs分泌IL-12pTo和IL-1B水平、下调IL-10的分泌水平。可因此得出推论:PL—PS能够通过调控DCs分泌Th1、Th2型细胞因子及趋化因子水平,诱导Th1型细胞免疫应答。

Abstract:To investigate the effects of polysaccharides isolated from the seeds of Plantago asiatica L. (PL-PS) on the secretion activity of bone marrow-derived dendritic cells (DCs). Bone marrow-derived dendritic cells were induced by 5-day-cuhure in RPMI-1640 medium supplemented with 10% heat-inactivated fetal calf serum, recombi- nant mouse granulocyte-macrophage colony-stimulating factor and interleukin-4 ,and then CD1 1c ~ cells isolated by MACS were cultured for 24 h in the presence of PL-PS at varying concentrations. The levels of IL-12p70, IL-10 and IL-1 β in culture supematant of DCs were detected by ELISA. Results showed that PL-PS up-regulated the secretion of IL-12p70 and IL-1β ,down-regulated the secretion of IL-10 in a dose-dependent manner at 25 -100μg/mL. The effects of PL-PS on the cytokine secretion of DCs suggested that PL-PS may play a role in inducing cellullar immu- nologic response.
