SPR光谱信号的一阶矩数据分析方法

摘    要:为了降低光源光强波动对表面等离子体共振(SPR)光谱信号的影响,提出一种基于幅值的SPR光谱信号动态基线一阶矩分析法.光源光强的波动会导致SPR光谱信号的幅值出现整体偏移,但SPR光谱信号的幅度变化较小,该方法的基线根据SPR光谱信号的最小幅值而动态调整.运用自行研制的SPR检测系统,测量了多种质量浓度的氯化钠和葡萄糖溶液得到光谱信号.采用固定基线法、固定宽度的动态基线法、基于面积的动态基线法和基于幅值的动态基线法等4种不同的一阶矩分析法得到光谱信号的共振像素.结果表明:动态基线法明显好于固定基线法,基于幅值的动态基线法其检出限(LOD)接近或者略好于其他2种动态基线法,且其在计算时间上明显少于另外两者,更适合于采样频率更高、实时性要求更强的SPR检测器.

Abstract:A first moment analysis method of dynamic baseline based on amplitude was proposed in order to overcome the influence of fluctuations of optical power on surface plasmon resonance (SPR) spectrum signals. The amplitude changes little while the values of SPR spectrum signals fluctuate because of the instability of optical power. This method adjusts the baseline dynamically according to the minimum value. Four different first moment analysis methods were used to obtain the resonance pixels by calculating the spectrum signals which were obtained by measuring NaCl and C6H12O6 solutions of various concentrations using a self-building SPR detection system. The resonance pixels of spectra were determined by the fixed baseline method, the fixed width based, the area based, and the amplitude based dynamic baseline method. The result shows that the dynamic baseline methods are significantly better than the fixed baseline method. The amplitude based dynamic baseline method is close to or less than the area based and fixed width based dynamic baseline methods on detection limit, and needs much less calculating time, so it is more suitable for SPR detectors which have high requirement for sampling rate and real-time.
