几丁质固定化纤维素酶降解壳聚糖的研究

摘    要:采用天然高分子聚合物几丁质作载体,以戊二醛为交联剂,对纤维素酶进行固定化,研究了戊二醛浓度、pH值、加酶量对纤维素酶固定化的影响,并采用固定化酶反应器对壳聚糖进行降解,再通过超滤膜装置进行分离,经液相凝胶色谱分析,降解产物平均分子量为2900,取得了很好的降解效果。同时对该反应器连续运行,测定该固定化酶的半衰期为60天,且降解性能保持稳定。

Abstract:Cellulase was immobilized on chitin by glutaraldehyde through cross link reaction in this work. The effects of glutaraldehyde concentration, pH, and the volumn of enzyme solution on the immobilized enzyme and the optimum conditions were determined. Furthermore, the immobilized enzyme column was used to hydrolyze chitosan, and the product of hydrolysis was separated by a ultrafiltration system. The average molecule weight of the product after separation was 2900 by GPC, which showed excellent hydrolysis ability. The immobilized enzyme bioreactor run stably and effectively, and the remaining of immobilized enzyme activity was 58% after continual work for 60 days.
