摘    要:目的观察二乙烯亚胺(BEI)对流感病毒的灭活效果。方法分别使用甲醛和BEI对流感病毒进行灭活试验,通过血凝试验检测二者灭活效果,并确定各自最佳灭活时间。用灭活的病毒液免疫小鼠,检测其抗原性,并用RT-PCR方法分析灭活病毒基因组的完整性。以裂解剂TritonX-100裂解病毒后,分别加入甲醛和BEI灭活,采用单向免疫扩散法检测灭活不同时间病毒液的HA含量。结果BEI灭活36h可将病毒完全灭活;BEI灭活的病毒免疫小鼠较甲醛灭活病毒产生较高的血凝抑制抗体;BEI灭活病毒液经RT-PCR,不能扩增出病毒基因;BEI灭活病毒的HA平均含量比甲醛灭活病毒的HA平均含量略高。结论BEI在灭活流感病毒感染性的同时,能够灭活病毒基因,并较好地保持病毒的抗原性。

Inactivation of Influenza Virus with Binary Ethylenimine
Abstract:Objective To observe the inactivation effect of binary ethylenimine (BEI) on influenza virus.Methods Inactivate influenza virus with formaldehyde and BEI respectively,determine the inactivation effect by HI test and optimize the time for inactivation.Determine the antigenicity of inactivated virus by immunization of mice.Analyze the integrity of genome of inactivated virus by RT-PCR.Lyze influenza virus with Triton X-100 and inactivate with formaldehyde and BEI respectively.Determine the HA content in virus liquid inactivated for various hours.Results Influenza virus was completely inactivated 36 h after treatment with BEI.The HI antibody titer of mice immunized with influenza virus inactivated by BEI was significantly higher than that by formaldehyde.No viral gene was amplified by RT-PCR from the virus liquid inactivated with BEI.After treatment with Triton X-100,the mean HA content in virus liquid inactivated with BEI was slightly higher than that with formaldehyde.Conclusion BEI inactivated both infectivity and gene of influenza virus.However,the antigenicity of inactivated virus was maintained.
