糖化酶As3.4309的固定化方法研究

摘    要:用蟹壳、明胶和卡拉胶做载体,采用交联法和包埋法对糖化酶As3.4309进行固定化。以淀粉溶液做底物,在相同条件下,对各种固定化糖化酶进行活力测定。实验结果表明:用卡拉胶包埋的固定化糖化酶保留了最高、的酶活力;而酶活力半衰期最长的是用蟹壳与戌二醛交联的方法制得的固定化糖化酶,对所获得的结果进行了分析,从而为固定化酶的进一步研究提供了方法上的依据。

Abstract:Chitin, gelatin and k-carrageenin are adopted as support matrix to immobilize glucoamylase As3. 4309 using entrapment and cross-linking methods. 1% starch solution is used as substrate to detect the activity of the immobilized glucoamylase. Under the same conditions of reaction, the experimental results showed that the activity of enzyme immobilized by entrapment to k-carrageenin remained the highest level, and the enzyme immobilized by cross-linking to chitin with pentanedial as cross-linking reagent was the most stable.
