苦荞糠皮总黄酮的抗氧化活性及免疫调节活性

摘    要:目的:评价苦荞糠皮总黄酮提取物的抗氧化及免疫活性,为其综合利用奠定基础。方法:测定苦荞糠皮黄酮对DPPH.、O2-.和.OH的清除能力和还原力,评价其抗氧化活性。测定不同剂量的苦荞糠皮黄酮提取物对正常小鼠的脾脏指数、吞噬细胞吞噬能力、迟发型变态反应、溶血反应的影响,评价其免疫调节活性。结果:在试验浓度范围苦荞糠皮总黄酮提取物对DPPH.、O2-.及.OH的清除率分别达到94.95%,86.4%,84.72%,其还原力随着浓度的提高显著增强。中、高剂量组均可极显著提高小鼠的脾脏指数(P<0.01);与对照组相比,对小鼠吞噬细胞的吞噬能力,迟发型变态反应以及血清溶血素水平的影响,中剂量组达显著水平(P<0.05),高剂量组达极显著水平(P<0.01)。结论:苦荞糠皮总黄酮提取物具有较强的抗氧化活性,对正常小鼠的非特异性免疫、细胞免疫和体液免疫功能都有促进作用,并具剂量效应。

Abstract:Objective:Investigate the anti-oxidation and immuno-modulatory efficacy of the flavonoids extracted from tartary buckwheat chaff for its further applications.Methods:the anti-oxidation activity was evaluated by determine the ability to scavenge the DPPH.,O2-.and.OH,the reducing ability was also determined.The effects on spleen weight,phagocytosis,delayed type hypersensitivity(DTH),sheep red blood cell(SRBC) lysis were investigated to evaluate the immunomodullatory efficacy of the extract.Results:In the tested range of concentration,the scavenge rate for DPPH.,O2-.and.OH reached to 94.95%,86.4%,84.72% respectively.The reducing power increased with the increase of the concentration used.For the normal mice,their spleen index were significantly increased by the intragastrically adminis tered medium and high dose of the extract(P<0.01).In comparison to the control group,both medium dose group(P< 0.05)and high dose group(P<0.01) significantly increased enhanced the phagocytic ability of phagocytes,DTH and serum hemolysin level.Conclusion:Flavonoids extracted from tartary buckwheat chaff exhibited strong antioxidant activities and enhanced the non-specific immunity,cellular and humoral immunity of normal mice.It also showed a dosage dependant effect.
