桦褐孔菌乙醇提取物对小鼠体内外抗氧化作用研究

摘    要:目的:研究桦褐孔菌菌丝体乙醇提取物的抗氧化活性。方法:采用正交试验方法,设置不同因素水平,确定最佳提取工艺。使用丙二醛(MDA)测定试剂盒测定桦褐孔菌菌丝体乙醇提取物对小鼠体内外MDA含量的影响,通过测定脂肪氧合酶和清除DPPH自由基等对桦褐孔菌菌丝体乙醇提取物的抗氧化活性进行研究。结果:乙醇提取物的最佳提取方案是:60%乙醇,80℃,料液比1:20,提取3h,此时提取率为12.9%。0.5~5.0mg/mL的乙醇粗提物具有较强的体内外抗氧化能力,可减少小鼠肝匀浆MDA的生成,抑制H2O2诱导的小鼠红细胞溶血。5.0mg/mL乙醇提取物对DPPH的清除率高达89.73%,对脂氧合酶的酶活性抑制率超过50%,并具有一定的剂量-效应关系。结论:本研究结果为研发桦褐孔菌菌丝体抗氧化、降血脂等功能奠定了理论基础。

Abstract:Objective:To investigate the antioxidant activity of ethanol crude extracts from mycelia of Inonotus Obliquus.Methods:Adopting orthogonal test methods,the technology of extracting the antioxidants from the mycelia of lnonotus Obliquus was studied and the antioxidant activities of the extracting solution under the optimal technology were analyzed.Results:The results showed that the optimal parameters of the extracting technology were that the concentration of ethanol was 60%;the extracting temperature was 80 ℃;the ratio of the sample to extracting solution was 1:20 and the extracting time was 3 h.The inhibiting rate was 12.9%.0.5~5.0 mg/mL ethanol extracts have strong antioxidant ability in vivo and in vitro.It could decrease MDA formation of mice liver homogenate and inhibit the hemlysis of mice red blood cell induced by H2 O2.5.0 mg/mL ethanol extracts,radical scavenging of DPPH was 89.73%,inhibit on lipoxdase activity was more than 50%.And with a dose-response relationship.Conclusion:The present study provide the theoretical basis for the further research and development of the antioxidant activity and hypolipidemic effect of mycelia of Inonotus Obliquus.
