聚乙二醇修饰纤维素酶的性能研究

摘    要:以单甲氧基聚乙二醇丙醛(mPEG-ALD 5KD)为修饰剂,对纤维素酶进行化学修饰,得到修饰纤维素酶(mPEG-Cell)。用mPEG-Cell降解微晶纤维素,以所得还原糖量为标准(DNS比色法测定),考察各种因素对其催化性能的影响,确定最佳修饰条件。结果表明:在0.1mol·L-1 pH=4.8的柠檬酸-柠檬酸钠的缓冲溶液中,纤维素酶质量浓度为5mg·mL-1,m(mPEG-ALD)∶m(酶)=1∶1,35℃修饰24h,所得到的mPEG-Cell具有最佳的催化性能,在50℃下降解微晶纤维素,24h后可得还原糖量达到2.58mg·mL-1,比同条件下天然酶降解微晶纤维素得到的还原糖量多0.53mg·mL-1,即多26%。

Abstract:The modified cellulase(mPEG-Cell)was obtained by using polyethylene glycol-aldehyde(mPEG-ALD) as modifier.The mPEG-Cell was used to degrade microcrystalline cellulose and the effects of various factors on its catalytic properties were investigated based on the amount of obtained reducing sugar(by DNS colorimetric analysis).The obtained optimum conditions were using 0.1M citric acid-sodium citrate as buffer(pH=4.8),concentration of cellulase 5 mg·mL-1,m(mPEG-ALD)∶ m(Cellulase)=1∶1,temperature 35 ℃ and time 24 h.The mPEG-Cell obtained under above conditions was used to degrade microcrystalline cellulose at 50 ℃ for 24 h,the amount of reducing sugar could reach 2.58 mg·mL-1,which was 0.53 mg·mL-1(26%) more than that obtained by using natural cellulase.
