酶法辅助超声波提取火龙果多糖及其抗菌活性

摘    要:采用单因素和响应曲面法优化酶法辅助超声波提取火龙果多糖的最佳工艺条件,并研究火龙果多糖的抗菌活性。结果表明:酶法辅助超声波提取火龙果多糖最佳条件是辅助酶为纤维素酶,酶用量0.1200 g,液料比20∶1,提取时间50 min,提取温度65℃,多糖得率可达16.64%;抑菌实验表明火龙果多糖的抗菌活性强弱顺序为大肠杆菌>金黄色葡萄球菌>啤酒酵母>黑曲霉>枯草芽孢杆菌。最小抑菌质量浓度(MIC)在5~30 mg/m L之间。

Abstract:The optimum processing conditions of enzyme- assisted extraction of polysaccharides from Hylocereus undatus were studied by single factor experiments and response surface. The antibacterial activity of Hylocereus undatus polysaccharose was evaluated. Results showed that the optimum process parameters were cellulose enzyme,the dosage of Enzyme was 0.1200 g,liquid- solid ratio was 20∶1,extraction at 65 ℃ for 50 min.The yield of Hylocereus undatus polysaccharides under the optimized conditions was 16.64%.Antibacterial results showed that the polysaccharide of Hylocereus undatus had good antibacterial activity,and best against Escherichia coli,followed by Staphylococcus aureus,Saccharomyces carlsbergensis,Aspergillus niger,antibacterial activity against Bacillus subtilis had the lowest activity. The minimal inhibitory concentrations( MIC) varied from 5 mg / m L to30 mg / m L.
