东宫泉对黑素细胞增殖及其酪氨酸酶活性的影响

目的探讨东宫泉对黑素细胞增殖及其酪氨酸酶活性的影响。方法体外培养小鼠B16黑素瘤细胞,倒置显微镜下观察不同浓度东宫泉(将50倍浓缩液0、2、4、8、16、32、64倍稀释)对B16黑素瘤细胞形态的影响,MTT法检测不同浓度东宫泉对细胞增殖的影响,蘑菇酪氨酸酶多巴速率氧化法测定不同浓度东宫泉对B16黑素瘤细胞酪氨酸酶活性的影响。结果经16倍稀释的东宫泉处理48h的B16黑素瘤细胞生长旺盛;东宫泉对B16黑素瘤细胞具有高浓度抑制、中低浓度激活的作用,8倍和16倍稀释为最佳激活浓度(P<0.05);东宫泉对酪氨酸酶亦具有高浓度抑制、低浓度激活的作用。结论东宫泉对黑素细胞增殖和酪氨酸酶均具有双向调节作用。

Effect of Orient House-Chuen,A Concentrate of Deep Underground Mineral Spring Water,on Proliferation and Tyrosinase Activity of Melanocytes

Objective To investigate the effect of Orient House-Chuen,a concentrate of deep underground mineral spring water,on the proliferation and tyrosinase activity of melanocytes.Method Murine melanoma B16 cells were cultured in vitro and treated with Orient House-Chuen at various concentrations(concentrated by 50 folds then diluted by 0,2,4,8,16,32 and 64 folds respectively),then observed for morphology under invert microscope and determined for proliferation level by MTT method and for tyrosinase activity by Dopamine oxidation rate method.Result The B16 cells treated with Orient House-Chuen at dilution of 1:16 for 48 h grew well as compared with those at other dilutions.Orient House-Chuen at high concentrations inhibited while that at low concentrations activated B16 cells as well as their tyrosinase activity.The optimal dilutions of Orient House-Chuen for activation were 1:8 and 1:16,at which the activation effects showed significant differences with those in control group(P < 0.05).Conclusion Orient House-Chuen showed dual regulatory effect on proliferation and tyrosinase activity of melanocytes.
