色氨酸在自制人血清白蛋白手性柱上的拆分及手性识别机理研究

摘    要:人血清白蛋白与三氯聚氰活化的氨丙基硅胶反应,制得人血清白蛋白键合手性固定相。反相模式下,色氨酸在该手性固定相上获得理想的拆分,分离因子可达3.51,分离度达5.49。探讨了流动相p H值、有机修饰剂、柱温等对手性拆分的影响。通过前沿分析法对色谱保留机理进行了探讨。

Enantioseparation of Tryptophan on Human Serum Albumin-immobilized Chiral Stationary Phase and Study on the Mechanism of Chiral Recognition
Abstract:Human serum albumin(HSA)based chiral stationary phase(CSP)was conveniently prepared by HSA and 2,4, 6-trichloro-1,3,5-triazine activated aminopropyl silica gel. Tryptophan was enantioseparated successfully on the as-pre?pared CSP under the reverse-phase mode. The separation factor and the resolution reached 3.51 and 5.49,respectively. The effects of pH value of mobile phase,type and content of organic modifier,and column temperature on the enantiosepa?ration of tryptophan were investigated. The mechanism of chiral recognition was studied further with frontal analysis method.
