核茎点霉发酵产纤维素酶发酵条件优化

摘    要:前期的单因素试验研究表明,在核茎点霉(Phoma tputaminum)LYYZ90-19发酵产纤维素酶过程中,温度、pH值及接种量是影响产酶量的关键因素。在此基础上,运用响应面分析法,以酶活力值为响应值,建立酶活力值与温度、prI值及接种量这3个关键因子的二次多项式回归模型,以Design Expert软件对模型进行分析并优化。研究表明,核茎点霉发酵产纤维素酶最佳工艺条件为:温度27.4℃、pH6.88、接种量6.75%。在该工业条件下,发酵液酶活为(18.21±0-3)U/mL。

Abstract:Previous single factor test results indicated that temperature,pH value and inoculating quantity were the key factors influencing cellulase yield of Phoma putaminum LYYZ90-19.Based on previous study,response surface method was applied to construct two degree polynomial regression models of the three key factors with cellulase activity as response values,and Design Expert software was used to optimize these models.The results showed that the optimal conditions were as follows: temperature,pH and inoculating quantity were 27.4 ℃,6.88 and 6.75 % respectively.Under the optimum conditions,the produced cellulase activity was as high as(18.21±0.3) U/mL.
