杂合酶的研究现状与发展前景

摘    要:近年来,随着人们对环境和能源问题的关注,以酶为催化剂的绿色生物催化技术越来越受到重视,而酶的催化能力是影响酶能否进行工业化应用的重要因素,同时,因催化不同反应的需要,新酶开发显得十分迫切。杂合酶技术是基于已有酶资源来进行新酶的设计和开发的技术,其具有独特的优点,尤其是目前酶的表达克隆、分子筛选、人工进化、DNA序列改组和体外突变等技术已成为常规技术,为杂合酶的开发及生产奠定了基础。本文对杂合酶的构建策略、构建方法及发展前景作一综述,为其应用和开发提供参考。

Current status and prospect of research on hybrid enzymes
Abstract:Along with the emphasis on energy and environmental issues,the green biocatalyst technology based on enzyme catalysis has been paid more and more attention in recent years.Catalytic ability is an important influencing factor on the industrial application of an enzyme.It is urgent to develop novel enzymes for various industrial processes.As a novel technique based on development from original enzymes by design and modification,hybrid enzymes share the benefits of original enzymes and show new properties.Several techniques,such as cloning and expression of enzyme,molecular screening,artificial evolution,DNA shuffling and in vitro mutation,have become routine techniques,which lay a foundation of development and production of hybrid enzymes.This paper reviews the strategy and method for construction as well as the prospect of hybrid enzymes in order to provide reference for application and development of the enzymes.
