摘    要:建立了反相高效液相色谱法测定百合中氨基酸含量的方法。采用HypersilODS2C18色谱柱(4.6mm×250mm,5μm),流动相A乙酸钠+四氢呋喃+三乙胺;流动相B:乙酸钠+甲醇+乙腈,柱温35℃,检测波长330nm。梯度洗脱程序为(t(梯度洗脱时间),B(流动相B)%)为(22,40);(23,60);(23.6,60);(26,65);(26.1,70);(26.6,80);(31.9,90);(32,95);(39,100);(40,0)。测定结果表明,在此实验条件下能使百合中13种氨基酸在40min内得到较好的分离,特别是脯氨酸能得到很好的分离。除trp、gly外,其他氨基酸具有较好的线性(相关系数为0.8985~0.9997)和重复性(相对标准偏差≤5.0%)。该方法简便实用,实际样品分析结果令人满意。

Abstract:A RP-HPLC method for the determination of amino acid in Lilium was established. A ODS2 Hypersil C18 column(4.6 minx250 mm, 5 μm)was used with solution A of NaAc-triethylamine-THFand B of NaAc-acetonitrile-methanol as mobile phase. Column temperature 35 ℃ and detection at 330 nm. The Volumn fraction of B mobile phase was programmed linearly in sections as follows: 0-22 rain, from 0 %-40 %; 22-23 rain, from 40%-60 %; 23-23.6 rain, from 60 %-65 %; 23.6-26.6 rain, from 65 %-80 %; 26.6-32 rain, from 80 %-95 %. The results showed that 13 kinds of amino acids were obtained in Lilium especially Pro. Amino and good linear relationship(r^2=0.8985-0.9997) and repeatability (RSD≤5.0 %). The method is simple and practical and the samples were determined with satisfaction.
