氢离子敏场效应管酶免疫传感器的研制

摘    要:介绍了以氢离子敏场效应管为基体电极,以碱性磷酸酶抗碱性磷酸酶抗体为例,在方法学上实现了电化学测量的一种酶免疫传感器.本生物传感器的响应时间达18s,标准曲线线性范围为7.8×10(-2)μg/ml~0.63μg/ml。另外,还讨论了适宜测量条件、抗体膜的寿命、抗原抗体复合物的寿命、抗原抗体复合物的分离及用不同抗体膜测量的重复性等。

Abstract:An enzyme immunosensor based on a pair of hydrogen ion sensitive field effect transistors (H+-ISFET) for the determination of alkaline phocphatase anti-alkaline-phosphatase antibody (APAAP) is reported. A linear relationship was obtained between the respoase peak and the logarithm of APAAP concentration from 7.8×10(-2)μg/ml to 0.63μg/ml. The response the time was only 18s.The appropriate condition for the measurement, the reversibility of the measurements with different membranes and the methods to dissociate the GAM-APAAP immunocomplexes are also discussed.
