百蕊草中总黄酮的提取工艺研究

用正交试验法优选百蕊草中总黄酮的提取工艺．以50g原药材的总黄酮提取量为评价指标，选择提取用水量（倍）、提取时间（h）、提取次数（次）、醇沉浓度（％）为考察因素，采用正交试验法L9（3^4）确定百蕊草中总黄酮的最佳提取工艺，采用分光光度法对干浸膏中总黄酮的含量进行了测定，检测波长274nm．最佳提取工艺条件为提取所用水量为15倍，提取次数为2次，回流提取时间为1h，醇沉浓度为75％，芦丁在2．06—12．22μg/Ll浓度范围之内呈良好的线性关系．平均回收率为99．57％，RSD为0．83％．总黄酮正交提取工艺的研究，结果比较可靠，含量测定方法简便、准确，可用于百蕊草干浸膏中总黄酮的含量测定．

Study on extraction process of total flavonoids in thesium chinense turcz

To study the optimum extraction process of total flavonoids in Thesium chinense turcz.The extraction of total flavonoids was chosen as the assessment index.Volume of water,extracting time and times,alcohol concentration were the study factors.The extraction process was optimized with the orthogonal design using spectrophotometric to determine total flavonoids content at 274nm wavelength.Results:The optimum ethanol extracting process conditions were as follows:adding 15 fold of water extraction two times,heating for 1 hours with 75% concentration alcohol.The rutin standard sample showed a good linear relationship at the range of 2.06 to 12.22g/mL.The average recovery was 99.57% and RSD was 0.83%.Conclusion The optimum extraction process of total flavonoids in thesium chinense turcz was reliable,easy and accurate which can be used for the total flavonoids extraction of thesium chinense turcz.
