大孔树脂分离纯化沙芥黄酮的工艺研究

摘    要:用大孔吸附树脂分离纯化沙芥总黄酮,比较了7种大孔树脂对沙芥总黄酮的静态吸附动力学特性,优选出D-4020型大孔吸附树脂分离纯化沙芥总黄酮,并对其进行动态吸附实验。结果表明,D-4020纯化沙芥总黄酮的最佳工艺参数为:上样液浓度0.4 mg/mL,pH值5,上样流速2 mL/min;使用4BV用量95%的乙醇作为洗脱剂,洗脱流速为2 mL/min。采用该工艺分离纯化沙芥总黄酮含量达40.91%。

Abstract:A method of separation and purification of flavonoids from Pugionium cornutum(L.) Gaerth.was studied.The suitable resin D-4020 was choosed by static dynamic adsorption experiments of seven macroporous resin.The adsorption experiment of D-4020 resin was done to determine the best condition.The optimal purification conditions were as follows:the sample solution concentration 0.4 mg/mL,pH 5,the sample flow rate at 2 mL/min;the elution solvent 4BV of 95 % ethanol,the flow rate at 2 mL/min.Under above condition,the purity of flavonoids could reach to 40.91%.
