反相色谱法测定银杏叶中的芦丁

摘    要:采用反相高效液相色谱法测定银杏叶中芦丁的含量。样品经乙醇回流提取、浓缩后,用Eclipse SB-C18(5μm,4.6mm×150mm)色谱柱分离样品,在检测器波长360nm,甲醇∶1.5%醋酸(体积比40∶60)为流动相,流速0.8mL/min,柱温30℃的条件下测定。该方法分离效果好,线性相关系数r=0.99987,平均回收率为98.69%,相对标准偏差为7.8%(n=5)。

Abstract:A new method was developed for the determination of rutin by reversed phase-high performance liquid chromatography.The effects of the kinds of extractant,the kinds and proportion of mobile phase,column temperature,flow rate of mobile phase,and ultraviolet detection wavelength were optimized.The analysis was performed on Eclipse SB-C18(5 μm,4.6 mm×150 mm) column with the mobile phase contained methanol-1.5% acetic acid(40∶60).The flow rate was 0.8 mL/min and the detection wavelength was set at 360 nm.The temperature of column was 30℃ and the injected volume was 20 μL.Under this condition,the linear coefficient was 0.99987 and the spiking average recovery was 98.69%.The relative standard deviations of the method was 7.8%(n=5).The results show that the method is reliable,fast and sensitive on the real samples.
