嗜热真菌发酵产木聚糖酶培养基的优化及部分酶学性质的研究

摘    要:采用响应面法对嗜热真菌发酵产木聚糖酶的培养基进行优化以及对该酶液的基本性质进行研究。首先对初始培养基中的8种影响因素进行Plackett—Burman设计,筛选出3种重要影响因素,即玉米芯、尿素和KH2PO4;再利用最陡爬坡实验为中心组合实验确定最大响应区间,最后经响应面分析获得最优化结果:玉米芯浓度为4.570%(w/v),尿素浓度为1.797%(w/v),KH2PO4浓度为0.497%(w/v),酶活提高了18%。该酶对玉米芯为底物的专一性较好,最适酶促反应温度为65℃且在该温度下较稳定,最适反应pH为6.8,并且pH稳定性较好。

Abstract:Fermentation medium of thermophilic fungal for xylanase production was optimized by response surface method and the basic properties of the enzyme solution were studied.
