乳香药材鉴别和含量测定方法研究

目的制定乳香药材的鉴别和含量测定方法,为药用植物资源的开发利用提供科学依据。方法采用薄层色谱法(TLC)鉴别乳香药材中主要有效成分11-羰基-β-乙酰乳香酸;采用高效液相色谱法(HPLC)测定其含量并制定含量限度,色谱柱为XTerraMS C18(3.9 mm×150 mm,5μm),流动相为乙腈-0.1%冰醋酸溶液(80:20),检测波长250 nm。结果乳香药材中11-羰基-β-乙酰乳香酸的TLC鉴别特征明显,专属性强;乳香药材中11-羰基-β-乙酰乳香酸的含量测定线性范围为0.256~5.12μg(r=1.000),平均回收率(n=6)为99.43%(RSD=1.21%),规定乳香药材中11-羰基-β-乙酰乳香酸的含量不得低于2.0%。结论本方法能快速、简单、准确地对乳香药材中有效成分11-羰基-β-乙酰乳香酸进行定性定量测定。

Study on Identification and Determination Methods of Boswellia carterii Birdw.

Objective To provide scientific basis for the utilization and development of Boswellia carterii Birdw. by establishment of identification and determination methods. Methods The bioactive constituent acetyl-11-keto-β boswellic acid in Boswellia carterii Birdw. was identified by TLC. Its content was determined by HPLC. HPLC was performed on C18 XTerra MS column, using acetonitrile-0.1% acetic acid（80：20） as mobile phase, the detection wavelength was 250 nm. Results The linear range of acetyl-11-keto-β-boswellic acid was 0.256-5.12 μg（r=1.000）, The average recovery was 99.43 %, with RSD of 1.21%（n=6）. The content of acetyl- 11-keto-p-boswellic acid in Boswellia carterii Birdw. should be not less than 2.0 %. Conclusion The established methods are simple and accurate, and can be used for the quality control ofBoswellia carterii Birdw.
