摘    要:目的优化水提醇沉法提取黄芪多糖的工艺条件,并比较不同产地黄芪药材中的多糖含量。方法在单因素考察提取黄芪药材多糖条件的基础上,采用正交试验设计法,以黄芪药材多糖含量为指标,确定多糖回流提取的最佳工艺;并采用苯酚-硫酸法测定不同产地黄芪药材的多糖含量。结果建立了黄芪药材多糖的最佳提取工艺:料液比1:20;醇沉浓度70%;回流提取3次,每次50 min。收集的9个产地黄芪药材的多糖含量为63~104 mg/g。其中,山东平邑产黄芪的多糖含量相对较高,辽宁义县产黄芪多糖含量相对较低。结论优化的提取工艺提取多糖效果好,稳定可行;不同产地黄芪药材中多糖含量有一定差异。本研究为黄芪多糖的进一步研究和黄芪药材的质量评价提供了可靠的实验依据。

Abstract:Objective To optimize the extraction technology of polysaccharides in Astragali radix and to determine the contents of polysaccharides from the medicinal materials of different habitats. Methods The optimized extraction technology was investigated by orthogonal test with the content of polysaccharides as the index which was determined by phenol-sulfuric acid method, and the contents of polysaccharides from Astragali radix of different habitats were determined. Results The best extraction condition of polysaccharides in Astragali radix was as follows: the powder of Astragali radix was extracted for 50 minutes with 20 times of water by reflux for 3 times, and the polysaccharides extracted was deposited with 70 % ethanol. The contents of polysaccharides from Astragali radix of 9 habitats were between 63-104 mg/g. The content of polysaccharides is highest in Astragali radix of Shandong Pingyi and lowest in Astragali radix of Liaoning Yixian. Conclusion The contents of polysaccharides in Astragali radix from different habitats are different. The optimum extraction condition can be stable and reliable, which can provide an experimental basis for further study of polysaccharides and quality evaluation of Astragali radix.
