青霉菌FSWY_3固体发酵研究

摘    要:筛选了一株高产木聚糖酶及纤维素酶的青霉(Penicillium)菌株FSWY3,并研究了此青霉菌株在固体培养基中的发酵条件。该菌株的最佳培养条件为,起始pH为4.6;发酵温度为29℃,每克固体培养基的接种量为0.5mL孢子悬液(孢子悬液的浓度为4.7×107个/mL),玉米芯与麦麸的质量比为7∶3,尿素为氮源,发酵时间为3d。木聚糖酶活可达4236U/g固体培养基,纤维素酶活可达1895U/g固体培养基。将粗酶液在不同温度下保持1h,发现当温度超过50℃时,酶活损失较大,残余酶活力为52.7%,至60℃时,酶基本失活,酶活力损失达93.0%。青霉FSWY3以农林废弃物玉米芯、麦麸为营养基料,能够产生具有很高活性的木聚糖酶和较高活性的纤维素酶,用其发酵曲作为饲料添加剂,既可提高饲料利用率,又减少了环境污染。

Studies on Ferment of Penicillium FSWY3 Using Solid State Fermentation
Abstract:Penicillium strain FSWY3 producing enzyme highly was isolated. The fermentation conditions of the strain in solid culture was studied. The optimal culture conditions were as follows: initial pH 4.6, temperature 29 ℃, per gram solid medium included 0.5 mL (spores concentration 4.7×107 )spore suspension inoculum, the rate of corncob and wheat bran is 7∶3, nitrogen source is carbamide, cultivation time is 3 days. The xylanase activity may reach as high as 4 236 U/g solid medium, and the cellulase activity could reach as high as 1 895 U/g solid medium. The remnant xylanase activity can be 52.7% when the temperature remained 50 ℃ for one hour, and when temperature reached 60 ℃, the loss of the xylanase activity would reach 93.0%. Penicillium FSWY3 can produce high activity xylanase and cellulase, if the production is used as feed additive, which will highly increase the using rate of feedstuff, and can reduce the environment pollution at the same time. 
