Abstract:A water soluble acryloyl chitosan(CS-AC)was synthesized through modifying chitosan(CS)with acryloyl chloride in methylsulfonic acid.Under UV exposure,a fast crosslinking CS-AC/DTT hydrogel was then prepared based on thiol-ene click chemistry using dithithitol(DTT)as crosslinker and I2959 as photoinitiator.Infrared spectroscopy(FTIR)and nuclear magnetic resonance(1H-NMR)results qualitatively and quantitatively proved the vinyl grafting rate of around 100%.Comparing with the gelation time over 40 min of self-crosslinking hydrogel CS-CC-CS,that of click chemistry hydrogel CS-AC/DTT was shortened within 10 min with a vinyl/thiol feed ratio of 1∶0.5.Characterization results demonstrated the gel strength of 5-10 KPa and excellent swelling rate up to 1415%.Furthermore,cytotoxicity and antibacterial tests of hydrogels exhibited excellent cytocompatibility towards NIH-3 T3 cells and antibacterial properties towards resistant staplylococcus aureus methicillin-resistant staplylococcus aureus(MRSA).The above results reveal the potential application in drug loading and antibiosis for the synthesized hydrogel materials.
