双水相体系萃取藤茶蛇葡萄素工艺的优化

摘    要:本文研究了聚乙二醇-硫酸铵双水相体系的组成对藤茶蛇葡萄素萃取率的影响。通过单因素实验研究了聚乙二醇相对分子质量、聚乙二醇质量分数、硫酸铵质量分数、pH和NaCl质量分数对萃取率的影响。其次通过Plackett-Burman设计筛选出对萃取率影响显著的三个因素,分别为聚乙二醇质量分数、硫酸铵质量分数、pH。利用响应面法对影响显著的因素进行进一步的优化。优化后的最佳萃取工艺为:聚乙二醇1000质量分数28%、硫酸铵质量分数11.2%、pH为6.0。在该条件下的理论萃取率为98.82%,经过工艺验证实际萃取率为98.53%,与预测值接近,比优化前萃取率(71%)提高了约17.53%,有利于蛇葡萄素的进一步分离纯化或应用。

Abstract:In this work,the effect of the composition of polyethylene glycol(PEG)-ammonium sulfate aqueous two-phase system on the extraction rate of ampelopsin from Ampelopsis grossedentata was studied.The effects of the molecular weight of PEG,the mass fraction of PEG,the mass fraction of ammonium sulfate,pH and the mass fraction of NaCl on the extraction rate were studied by single factor experiment.Secondly,three factors with significant influence on extraction rate were selected through Plackett-Burman design,namely,mass fraction of PEG,mass fraction of ammonium sulfate and pH.Response surface methodology was used to further optimize the influential factors.The optimum extraction conditions were:PEG1000 mass fraction 28%,ammonium sulfate mass fraction 11.2%,pH6.0.Under this condition,the theoretical extraction rate was 98.82%,and the actual extraction rate was 98.53%after process verification,which was close to the predicted value and 17.53%higher than the extraction rate before optimization(71%),which was conducive to the further separation,purification and application of ampelopsin.
