不同产地百合药材中薯蓣皂苷元及总皂苷元的含量测定

摘    要:采用高效液相-蒸发光散色检测器(HPLC-ELSD)测定不同产地百合中薯蓣皂苷元的含量,比较了不同产地百合药材中薯蓣皂苷元及总皂苷元的含量。以甲醇-水(91∶9)为流动相;流速为1.0 mL/min;柱温30℃;漂移管温度55℃,压力1.40 bar。采用紫外分光光度法测定百合中总皂苷元的含量,浓硫酸作为显色剂,在285 nm处测定吸光度值。结果显示:不同产地百合药材中薯蓣皂苷元和总皂苷元的含量存在差异,各药材总皂苷元的含量在0.386%~1.179%,其中以江苏产的百合含量较高。采用浓硫酸-无水乙醇法测定百合中总皂苷元的含量,简单、快速、准确、重现性好,符合含量测定建立的要求

Abstract:The contents of diosgenin and sapogein in Herba Bulbus Lilii derived from different areas with HPLC - ELSD method were compared, with a mixture of methanol(A) and water(B) (91 : 9) as the mobile phase at a flow rate of 1 mL/min, the column temperature at 30 ℃, drift tube temperature at 55 ℃, and pressure of nebulizer gas of 1. 40 bar. Spectmphotometer was used to determine the Sapogein levels with H2SO4 for colorimetric reaction and at 285 nm ul- trasonic treament. The content of diosgenin and sapogein and the total saongein levels in each sample in different are as were between 0.386% - 1. 179%, in which the total sapogeins in lilium brownii from Jiangsu was the hightest, method of ethanol - sulfuric acid the to determine total sapogenin content in Bulbus Lilii was simple and accurate with a good sensi- tivity, which can be used for quality control of the medicinal Bulbus Lilii.
