食品中磺胺类药物化学发光酶联免疫检测试剂盒的初步研究

在常规酶联免疫法的基础上,将磺胺类母核与牛血清白蛋白合成免疫抗原后免疫小鼠制得磺胺类单克隆抗体,并进一步优化抗体工作浓度、反应时间等实验参数,建立了一种简便、快速、精确的磺胺类残留一步式化学发光酶联免疫检测方法.结果表明该方法最佳检测范围为1.0～81.0 ng/mL,检测IC50达3.047 ng/mL,对猪肉的最低检测限为0.74 ng/g,可检出国际规定的SAs残留标准底限值以下的残留量.与常规ELISA方法相比,检测限和灵敏度基本一致,但反应时间缩短了60 min.证明采用一步式化学发光酶联免疫检测磺胺类残留可以大幅度缩减检测时间,符合快速检测的要求.

Preliminary Study on Chemiluminescent Enzyme Immunoassay Kit for Sulfonamides in Food

Based on conventional enzyme linked immunoassay（ELISA）,a convenient,rapid and accurate one-step sulfonamides（SAs） residue chemiluminescent enzyme immunoassay（CLEIA）detection method was established through obtaining monoclonal antibodies from immunizing mice which was injected with SAs-BSA coupling protein,and optimizing test parameters such as antibody concentration,reaction time.The results showed that the best detection range of this method was 1.0～81.0 ng/mL,the IC50 was 3.047 ng/mL,the detection limit was 0.74 ng/mL.Compared with conventional ELISA,the CLEIA method can not only increase the detection limit and sensitivity,but also shorten the reaction time by 60 minutes.CLEIA kit is more appropriate for rapid detection of the SAs residue in food.
