浙江新分离狂犬病病毒街毒株与国内外疫苗株G基因的比较分析

摘    要:目的对浙江新近分离的2株狂犬病病毒街毒株的G基因进行遗传学分析,并比较其与国内外使用的狂犬病疫苗株间的差异。方法以直接免疫荧光法和双抗体夹心ELISA法检测伤人犬脑组织,乳鼠接种分离病毒,RT-PCR扩增G基因片段,直接测序后进行遗传学分析。结果在2份伤人犬脑组织(编号LH和ZJCA1)中均检出狂犬病病毒,与其他浙江街毒株相比,G基因核苷酸和氨基酸序列同源性均为98.7%~99.2%。与当前国内外使用的多种疫苗株相比,与CTN疫苗株同源性最高,G基因核苷酸和氨基酸序列同源性分别为79.5%~86.1%和86.8%~94.1%。结论浙江新分离的2株狂犬病病毒街毒株属于基因1型,与当前使用的多种疫苗株相比,与CTN疫苗株属于同一分支,二者的亲缘关系最近。

Comparison of G Genes of Novel Rabies Street Virus Strains Isolated in Zhejiang Province, China and Other Vaccine Virus Strains
Abstract:Objective To compare the G genes of novel rabies street virus strain isolated in Zhejiang Province, China and other vaccine virus strain used in China and abroad. Methods The brain tissues of a dog biting 5 persons in Chun'an County, Zhejiang Province(ZJCA1)and a dog biting 29 persons in Taizhou City, Zhejiang Province(LH)were detected by direct IFA and double antibody sandwich ELISA, and virus strains were isolated by inoculation i.c. in suckling mice, from which G gene fragment was amplified by RT-PCR for direct sequencing and genetic analysis. Results Rabies virus was detected in both dog brain tissue samples ZJCA1 and LH, of which the homologies of nucleotide and amino acid sequences of G gene were 98. 7% ~ 99. 2% to those of other street virus strains isolated in Zhejiang Province, and 79. 5% ~ 86. 1% and 86. 8% ~ 94. 1% to those of CTN strain respectively. Conclusion The two novel rabies street virus strains isolated in Zhejiang Province, China were of genotype 1 and located in the same branch with CTN strain on phylogenetic tree.
