植物乳杆菌BLPC002产苯乳酸发酵培养基的优化

摘    要:以L-苯丙氨酸为底物,对植物乳杆菌BLPC002产苯乳酸的发酵培养基进行了优化。通过单因素试验,研究了不同碳源及其他因素对苯乳酸产量的影响,采用Plackett-Burman试验设计法,从7个影响因素中筛选出苯丙氨酸、葡萄糖和牛肉浸粉3个主要影响因素,通过最陡爬坡试验和响应面法(RSM)优化了培养基的配方。结果表明,最佳培养基配方为苯丙氨酸8.3 g/L、葡萄糖30.0 g/L、牛肉浸粉1.8 g/L,优化后植物乳杆菌BLPC002产苯乳酸产量达到1.67 g/L,是优化前(0.36 g/L)的4.6倍。

Abstract:With L-phenylalanine as substrate, the fermentation medium formula of Lactobacillus plantarum BLPC002 for phenyllactic acid production was optimized. By single factor experiments, the effects of different carbon sources and other factors on the yield of phenyllactic acid were investigated. Three most significant factors affecting phenyllactic acid production were selected from seven factors with Plackett-Burman design, which were phenylalanine, glucose and beef extract powder, the medium formula was optimized by steepest ascent experiments and response surface methodology. The results showed that the optimal medium formula was phenyllactic acid 8.3 g/L, glucose 30.0 g/L and beef extract powder 1.8 g/L, respectively. The yield of phenyllactic acid reached 1.67 g/L after optimization, which was 4.6 times higher than that of before optimization (0.36 g/L).
