枯草芽孢杆菌Z-3产木聚糖酶固态发酵工艺优化

摘    要:以木聚糖酶活力为响应值,通过单因素及响应面试验优化克氏原螯虾肠道来源枯草芽孢杆菌(Bacillus subtilis)Z-3产木聚糖酶的固态发酵工艺。结果表明,最优固态发酵工艺为培养时间3 d、培养温度37 ℃、初始pH值5.0、装料量30 g/250 mL,接种量2.5%,碳源为麸皮与玉米芯混合物,麸皮占比60%,氮源为氯化铵。在此最优发酵工艺条件下,木聚糖酶活力为3 459.58 U/g湿基,较未优化前(2 079.1 U/g湿基)提高66.40%。

Abstract:Using xylanase activity as the response value, the solid-state fermentation technology for xylanase production by Bacillus subtilis Z-3 isolated from the intestinal of Procambarus clarkii was optimized by single factor and response surface tests. The results showed that the optimal solid-state fermentation technology was culture time 3 d, temperature 37 ℃, initial pH value 5.0, loading volume 30 g/250 ml, inoculum 2.5%, bran and corn cob (bran 60%) as carbon source, and ammonium chloride as nitrogen source. Under the optimal fermentation conditions, the xylanase activity was 3 459.58 U/g wet basis, which was 66.40% higher than that before optimization (2 079.1 U/g wet basis).
