西马特罗单克隆抗体制备以及酶联免疫试剂盒的研究

摘    要:通过制备西马特罗半抗原,载体蛋白与之偶联合成免疫原,免疫原用于免疫动物,获得抗西马特罗单克隆抗体,制备能够检测猪尿中西马特罗残留量的酶联免疫试剂盒。结果表明,该试剂盒对猪尿中西马特罗的检测限为0.295 μg/L,半抑制浓度(IC50)值为0.870 μg/L,回收率范围为81.5%~103.5%,标准曲线线性范围为0.1~8.1 μg/L,精密度试验结果批内、批间的相对标准偏差(RSD)均<15%,表明该检测方法准确度和精密度高。该试剂盒特异性较好,于4 ℃条件下能够保存12个月,稳定性较好,可以用于猪尿中西马特罗德快速检测。

Abstract:Cimaterol hapten was prepared and coupled with carrier protein to synthesize an immunogen. The immunogen was used to immunize the animal to obtain an anti-cimaterol monoclonal antibody, which was used to prepare for enzyme linked immunosorbent assay kit for detecting cimaterol residues in swine urine. The results showed that the detection limit of the kit for cimaterol in swine urine was 0.295 μg/L, the IC50 (50% inhibition concentration) value was 0.870 μg/L, the recovery range was 81.5%-103.5%, the linear range of the standard curve was 0.1-8.1 μg/L, and the relative standard deviation (RSD) within and between batches of precision experiment results was less than 15%, indicating that the detection method had high accuracy and precision. The kit had good specificity and could be stored at 4 ℃ for 12 months with good stability, and could be used for rapid detection of cimaterol in swine urine.
