褐藻胶裂解酶酶学性质及酶解马尾藻工艺的响应面优化

摘    要:海洋细菌是褐藻胶裂解酶的重要来源。该试验以实验室制备的褐藻胶裂解酶粗酶液为研究对象,对其酶学性质及酶解工艺进行初步研究。结果表明,酶的最适反应条件为50 ℃、pH 8,在4~40 ℃、pH 5~9范围内酶活力较稳定;Ca2+、Mg2+和Fe2+等离子对该酶有促进作用,EDTA、Ba2+、Zn2+等有抑制作用。以孤囊马尾藻(Sargassum oligocystum)为试验原料,以褐藻寡糖含量为响应值,通过单因素试验和响应面法对马尾藻的固态酶解工艺条件进行优化,当含水率75%、温度40.7 ℃、酶解10.3 h时,寡糖含量达到(50.42±0.24) mg/g。该试验可为褐藻寡糖的酶解法规模制备提供理论依据与应用参考。

Abstract:Marine bacteria are the important source of alginate lyases.In this study,using the crude alginate lyase solution prepared in the laboratory as research object,the enzymatic characteristics and hydrolysis process were preliminarily studied,and the results showed that the optimal enzymatic reaction conditions of the lyases were 50℃and pH 8,and the lyases activity was stable in the range of 4-40℃and pH 5-9.The Ca^2+,Mg^2+and Fe^2+promoted the enzyme activity,but EDTA,Ba^2+ and Zn^2+ had inhibitory effects.Using Sargassum oligocystum as experimental material and oligosaccharide content as response value,the solid-state enzymatic hydrolysis process condition was optimized by single factor tests and response surface methodology.The oligosaccharide content reached(50.42±0.24)mg/g when Sargassum powder(water content 75%)was enzymatically hydrolyzed at 40.7℃ for 10.3 h.The study could provide theoretical basis and application reference for the enzymatic scale preparation of alginate oligosaccharide.
