摘    要:酶解玉米蛋白粉(蛋白含量为70%)制备血管紧张素转换酶(angiotensin converting enzyme,ACE)抑制肽,通过酶的筛选实验确定了AS.1398中性蛋白酶作为最佳水解酶,在此基础上,进行pH、温度、底物浓度、加酶量[E]∶[S]的单因素实验,并且确定4种因素的参数值进行L9(34)正交实验,采用体外检测ACE抑制率和肽得率为指标来确定最佳工艺条件。研究结果表明,选用AS.1398中性蛋白酶作为水解酶,水解时间在2h时,pH7.0,温度50℃,底物浓度5%,加酶量[E]∶[S]为1.5∶100,得到的最大ACE抑制率为85.65%,肽得率为58.64%。

Abstract:Enzymatic hydrolysis of corn protein powder(protein content 70%) to produce ACE inhibitory peptides,by the enzymatic screening test to determine the AS.1398 neutral protease enzyme as the best,on this basis,used the pH,temperature,substrate concentration,the amount of enzyme [E]∶[S] in the single-factor test and determined the four factors in L9(34) orthogonal test,through vitro study to detect ACE inhibition rate and yield of peptide as indicators to determine the optimum technology conditions. The result indicated that AS. 1398 neutral protease was the best hydrolytic enzyme,when the hydrolysis time in 2h,pH7.0,temperature 50℃,5% substrate concentration,the amount of enzyme [E]∶[S] 1.5∶100,and the maximum ACE inhibition rate was 85.65%,the yield of peptide was 58.64%.
