硼酸对L-阿拉伯糖异构酶催化合成D-塔格糖的影响

摘    要:基于硼酸与构型不同的单糖的络合能力不同,探讨了添加硼酸对L-阿拉伯糖异构酶催化D-半乳糖异构化为D-塔格糖的酶反应体系的影响,研究了pH、温度、硼酸加入量、加酶量等条件对硼酸作用的影响。结果表明:当反应条件为pH9.0,温度65℃,硼酸与底物的摩尔比1∶2,加酶量为0.39U/mL时,90g/L的底物D-半乳糖的转化率为50%,而不加硼酸时转化率为27%,表明硼酸对反应有很好的促进作用。

Abstract:Based on different affinities of boric acid to D-galactose and D-tagatose,an L-arabinose ismoerase mutant enzyme was used to catalyze the isomerization of D-galactose to D-tagatose with boric acid.The effect of pH,temperature,molar ratio of boric acid and the enzyme addition was studied.The results showed the optimum conditions for production of D-galactose were pH 9.0,65℃,1∶2 molar ratio of boric acid to D-galactose,the amounts of enzyme was 0.39U/mL.Under this condition,in 16h,the convertion was 50% when the concentration of D-galactose was 90g/L with boric acid,while 27% in the Tris-HCl buffer system without borate,which showed that boric acid had good impact on the production of D-tagatose.
