黑曲霉液体发酵桔粉产纤维素酶的研究

摘    要:以桔粉为原料,以黑曲霉(Aspergillus niger)为发酵菌株,采用液体发酵法生产纤维素酶。通过单因素试验考查了麸皮和蛋白胨的比例(C/N)、装液量及接种量3个因素对产酶的影响,并在此基础上,通过正交试验确定了发酵产酶的工艺条件为:添加桔粉80 g/L,麸皮和蛋白胨质量比为1∶2、250 mL三角瓶装30 mL Mandels氏营养液、接种量3 mL,于30℃下培养72 h,纤维素酶产量达到1885.71 U/g。

Abstract:With orange powder as raw materials and Aspergillus niger as production strains,cellulase was producted by liquid fermentation.Three important factors of ratio of bran and peptone(C/N),liquid volume in flask,and inoculums size on cellulose activities were researched respectively by single factor test.Based on it,the enzyme production conditions were optimized by orthogonal test.The optimum conditions of enzyme production were adding 80 g/L orange powder,1∶2 ratio of bran and peptone,adding 30 mL Mandels nutrient solution to 250 mL flask,3 mL inoculums size,culturing 72 h at the temperature of 30 ℃.Activities of cellulase producted on the above conditions were 1885.71 U/g.
