摘    要:目的:建立测定金银花中绿原酸含量的HPLC方法,并比较金银花花茎叶中绿原酸含量.方法:采用高效液相色谱法,色谱柱:Agilent Extend-C18(4.6 mm×150mm,5μm),流动相:乙腈-水-磷酸(13∶87∶0.4),流速1.0 mL/min,柱温25℃,检测波长为329 nm.结果:绿原酸在10.4~62.4 μg/mL内线性关系良好,平均回收率98.7%,RSD为1.74%.同时期的金银花花、茎、叶中绿原酸的含量分别为6.67%、5.43%、4.80%.结论:该方法精密、准确、快速、重现性好,可用来测定比较金银花花茎叶中绿原酸含量的方法,经HPLC法确定金银花花茎叶中均含有大量绿原酸,具有较高的开发利用价值.

Abstract:Objective: To establish the assay method for determining content of chlorogenic acid in the Flowers of Lonicerae Japonicae Flos, and and compare the chlorogenic acid content in the flower, leaves, stems during the same growth periods by HPLC. Methd: Agilent Extend-Cl8 column (4.6 mm×150 mm, 5 μm) was used with a mobile phase othcetonitrile - water - phosphate(13 : 87 : 0.4), at the flow rate of 1.0 mL.min-l, and detection wavelength of 329 nm, the column temperature of 25 ~C. Result: Chlorogenic acid showed a good linear relationship within 10.4-62.4 μg/mL. The average of coefficient of recovery was 98.7 %, RSD was 1. 74 %, and chlorogenic acid content in flower, stems and leaves was 6.67 %, 5.43 % and 4.80 %. Conclusion: This method was simple, accurate and reproducible for the determination of chlorogenic acid in Lonicerae Japonicae FIos. It's sure that the flowers,stems and leaves contains a lot of chlorogenic acid and there is development and utilization prospect for the leaves and stems of Lonicerae Japonicae Flos.
