半乳糖化温敏水凝胶的制备及其作为原代肝细胞培养载体的研究

摘    要:文章在前期工作基础之上,进一步研究半乳糖糖化温敏水凝胶P(NIPAAm-co-GAC)的生物相容性.通过原位胶原酶灌注法分离得到大鼠正常肝细胞,将其接种于P(NIPAAm-co-GAC)水凝胶表面,在未加任何生长因子的前提下研究肝细胞的增殖行为及细胞功能的表达情况,MTT实验结果表明含有肝细胞特异性吸附配体的P(NIPAAm-co-GAC)(r=0.4)水凝胶能够促进细胞粘附增殖,细胞受损伤程度低,细胞膜保持较好的完整性(LDH漏出量少)并降低了细胞的炎症反应(ALT、AST、ALT/AST比值低),不会引起细胞坏死,并且肝细胞能保持其合成分泌白蛋白、尿素的功能.研究正常肝细胞比研究肝肿瘤细胞或其它细胞系细胞具有更大的临床价值,为其进一步应用于生物人工肝领域提供充分的理论依据.

Abstract:Based on previous work, we further studied the compatibility of P(NIPAAm-co-GAC) thermo-sensitive hydrogels in this paper. Rats normal liver cells were separated by in situ collagenase perfusion. Ceils were inoculated onto P(NIPAAm-co-GAC) hydrogels and cultured without any growth factors. Cells proliferation and the function of protein syntheses were evaluated. The results of the MTT assay showed that P(NIPAAm-co-GAC)(r=0.4) hydrogels were fitted for cells adhesion. Cells with integrity membranes had low degree of damage (LDH leakage less), and cells with low degree of inflammatory response (ALT, AST, ALT/AST less), liver cells could keep their function of synthesis protein of albumin and urea at the same time. There was more clinical value of investigating normal liver cells than liver tumor cells line or other cells line, our work provided theoretical basis for further research of P(N1PAAm-co-GAC) thermo-sensitive hydrogels in bioartificial liver.
