阿达帕林的精制工艺研究

摘    要:在negishi偶联后处理步骤中采用EDTA-2Na水溶液洗涤以除去无机盐。以四氢呋喃-甲苯重结晶,采用保温析晶,干燥后于甲醇和去离子水中回流搅拌,前后两次80℃真空干燥。结果单一杂质:0.06%、总杂质:0.17%;重金属、炽灼残渣和溶剂残留均能符合规定。

Abstract:Aqueous solution of EDTA-2Na was introduced in the work-up procedures of negishi coupling.With controlling crystallization temperature,the crude product was recrystallized by tetrahydrofuran-toluene and stirred at refluxing temperature in methanol and deionized water solution,with both before and after vacuum drying in 80℃ and crystallization,respectively.The results showed that the largest impurity result was 0.06% ,the total impurities result was 0.17% ,and none of heavy metal,ignition residue and residual solvent was found to exceed quality limit.
