高效液相色谱荧光法测定调味品中罗丹明B的含量

摘    要:建立了固相萃取净化高效液相色谱荧光检测器测定调味品中罗丹明B的方法。样品中的罗丹明B用水或甲醇提取,Sep-pak C18小柱净化,用高效液相色谱荧光检测器测定,外标峰面积法定量。结果表明:罗丹明B在5.0~100.0 ng/mL范围内呈良好线性关系,相关系数为0.9985,3个水平(5.0、10.0、25.0μg/kg)添加罗丹明B的回收率为81.0%~93.7%,相对标准偏差为1.62%~3.08%(n=6),检出限为0.5μg/kg。此方法具有简单、快速、准确等优点,适用于调味品中罗丹明B的测定。

Abstract:Solid phase extraction followed by high performance liquid chromatography(HPLC) with fluorescence detector method was developed for the analysis of Rhodamine B in condiment.The separation of Rhodamine B from samples was extracted by water or methanol,and purified by Sep-pak C18 column,and determined by HPLC with fluorescence detector(FLD).The calibration curve of the method was linear over the range of 5.0 to 100.0 ng/mL with a correlation coefficient of 0.9985.The recoveries for Rhodamine B standard spiked in the range of 5.0 to 25.0 μg/kg were between 81.0 % to 93.7 % with a relative standard deviation(RSD) being between 1.62 % to 3.08 %(n=6).The detection limit was 0.5 μg/kg.Owing to advantages of good linearity,high precision and rapidity,this method is suitable for the determination of Rhodamine B in condiment.
