降酚酵母菌驯化筛选及其降酚性能

摘    要:目的研究酵母菌对苯酚的降解能力,确定不同条件对酵母菌降酚能力的影响,选育出性能优良的降酚酵母菌菌株.方法采用孟加拉红培养基分离酵母菌菌株,测定其对苯酚的降解率,并分析不同温度、pH值、振荡速度对苯酚降解率的影响.结果四株菌株对苯酚都有一定的降解能力,其中101B、863B菌株降解效果较好,苯酚质量浓度为500mg/L时,两菌株24h降解率都达到94%以上;苯酚质量浓度为1000mg/L、l500mg/L时,101B降解效率高于863B,分别为90.18%、86.44%;101B菌株的最适条件为pH值4~5、温度25~30℃、振荡速度150r/min.结论101B菌株对高浓度苯酚有较好的降解率,pH值、温度对菌株的降酚能力影响较大,适当提高振荡速度有利于苯酚的降解.

Abstract:This paper studies the phenol degradation ability and the influence of the yeast strains in different conditions, screening the phenol - degrading yeast strains with excellent performance. The Rose Bengal Me- dium was used to isolate the yeast strains, measure the degradation rate of phenol, and analyze the influence of degradation rate of phenol in different temperature, pH and oscillation speed. This four strains all can have degradation phenol, among them, 101B, 863B performance well, when the phenol concentration is 500 mg/L, the degradation rate of both the strains reaches 94% in 24 hours. When the phenol concentration is 1 000 mg/L, 1 500mg/L,the degradation rate of 101B is 90. 18% ,86.43% respectively. 101B works well when pH is 4 -5 ,temperature is 25 -30~C and the oscillation speed is 150r/min. 101B is the highest con- centration - tolerant in this four strains, pH and temperature have great effect on phenol - degrading yeasts and increasing the oscillation speed appropriately helps raise the degradation rate.
