海洋假单胞菌PT-8胞外硒多糖的制备

摘    要:研究了硒的添加量对海洋假单胞菌PT-8硒多糖产量的影响,考察了胞外硒多糖的流变性。对海洋假单胞菌进行发酵培养,确定Na2SeO3加入量为20μg/mL,培养6h后加硒,发酵时间为48h。富硒多糖通过海洋假单胞菌PT-8进行深层富硒发酵,得到富硒多糖产量为7.28g/L,硒质量分数为256.7mg/kg。流变性实验结果表明,在不同浓度、温度和剪切力条件下,多糖的黏度高于硒多糖黏度。多糖在pH为4时黏度达到最大,为96mPa·s;硒多糖在pH为5时黏度达到最大,为80mPa·s。初步确定该多糖为酸性多糖。

Abstract:The influence of selenium concentration on the enriched-selenium exopolysaccharides(Se-EPS)production by Pseudomonas PT-8 was studied. The yield of enriched-selenium exopolysaccharide was 7.28g/L,Se-enriched amount was 256.7mg/kg when Se-concentration was20μg/mL,Se-added time 6th h,shake culture 48h.EPS showed stronger rheological property than Se-EPS.The viscosity of EPS at pH 4was 96mPa·s,but the viscosity of Se-EPS was 80mPa·s.It is primarily confirmed that Se-EPS and EPS should be acidic polysaccharide.
