水质净化芽孢杆菌的筛选及培养条件优化

摘    要:从养殖池底的海泥筛选出两株具有净化水质的细菌T01、T02,对其进行生理生化鉴定,确定均为芽孢杆菌。对T01、T02及复合菌株(体积比1∶1)降解氨氮和亚硝酸盐氮能力进行测试,菌株T01、T02均具有降解氨氮和亚硝酸盐氮的能力。选择生长状况较好的T02进行培养条件优化,在葡萄糖10g/L、淀粉15g/L、蛋白胨5.0g/L、豆粕10g/L、磷酸氢二钾1.5g/L、磷酸二氢钾1.5g/L、硫酸镁1.0g/L、碳酸钙1.0g/L的培养基条件下活菌数达到最高,为1.54×109 cfu/mL。在接种量1%、培养温度30℃、转速180r/min、装液量2/5时,活菌数达到最高,为1.63×109 cfu/mL。

Abstract:Strains T01 and T02 with water purifying function were isolated from sea-mud in culture pond bottom and identified as Bacillus.Both strains were capable of degrading ammonia nitrogen and nitrite nitrogen.The number of living bacteria achieved the highest at 1.54×109 cfu/mL when the glucose was 10g/L,starch 15g/L,peptone 5g/L,soybean meal 10g/L,potassium hydrogen phosphate 1.5g/L,potassium dihydrogen phosphate 1.5g/L,magnesium sulfate 1g/L,calcium carbonate 1g/L.The optimum growth occurred at 1%inoculation amount in 2/5fluid volume and at30 ℃,180r/min,in which the highest cells was 1.63×109 cfu/mL.
