茎瘤芥叶叶绿素提取工艺研究

摘    要:以茎瘤芥叶为原料,采用冷冻干燥技术对叶片进行预处理后,采用单因素试验和正交试验研究不同的浸提剂、乙醇浓度、料液比、浸提温度和浸提时间对茎瘤芥叶叶绿素提取率的影响,并优化茎瘤芥叶叶绿素提取工艺.结果表明:有机溶剂提取茎瘤芥叶叶绿素优化工艺为料液比1∶50,提取溶剂为乙醇体积分数70%的乙醇/丙酮溶液,50℃水浴温度浸提3h,叶绿素提取率为1.08%.

Abstract:In this study,freeze drying technique was applied to pretreat the leaves of Brassica juncea var.tumida Tsen et Lee.The chlorophyll’s extracting ratio was effected by the different extractant,alcohol concentration,solid-liquid ratio,extracting temperature,extracting time.These studies’ methods were single factor test and orthogonal test.Meanwhile the extracting technology was optimized.The optimized extracting conditions were as follows: solid-liquid ratio of 1∶50;alcohol concentration of 70%(vol) alcohol /acetone solvent,50 ℃ waterbath extracting time of 3 h.The chlorophyll’s extracting ratio was 1.08%.
