双水相体系萃取粗状假丝酵母脂肪酶的研究

摘    要:通过比较不同无机盐与PEG组成双水相体系对粗状假丝酵母脂肪酶的萃取效果,系统地研究了脂肪酶在PEG/(NH4)2SO4双水相体系中的分配行为,考察了PEG分子量、PEG浓度、(NH4)2SO4浓度和体系pH对脂肪酶分配系数和萃取率的影响,并通过响应面分析法进一步优化了萃取条件。结果表明最佳萃取条件为:PEG2000浓度18·3%,(NH)SO浓度17·4%,pH8·0,室温条件下其分配系数可达到13·3,纯化倍数达到1·53,萃取率达到92·2%。

Abstract:The extraction of Candida valida lipase by aqueous two-phase system was studied.The effects of salts,the molecular weight and concentration of polyethylene glycol (PEG), the concentration of( NH_4)_2SO_4 and pH value on the distribution coefficient K, yield Sf were examined. Response surface methodology was employed to further analyze and optimize the extraction of Candida valida lipase.The results showed that the optimum extraction conditions were PEG2000 18.3% (m/m), (NH,)2SO4 17.4% (m/m) and pH at 8.0.Under these conditions, the distribution coefficient, purification factor and yield of lipase could reach to 13.3,1.53 and 92.2%, respectively.
