白灵菇菌丝体多糖的分离纯化及理化性质研究

摘    要:研究白灵菇菌丝体粗多糖的分离纯化技术,并对得到的多糖进行纯度鉴定及理化性质研究。结果表明,白灵菇菌丝体多糖(PNMP)经DEAE-Cellulose52和Sephadex G-200柱层析后得到单一组分PNMPⅢ-a,经分析鉴定,PNMPⅢ-a为不含双糖、糖醛酸和核酸的非淀粉类的均一多糖;其相对分子质量为15213Da。PNMPⅢ-a完全酸水解后,经高效液相色谱分析确定其糖基的组成为葡萄糖(Glc)、鼠李糖(Rha)、木糖(Xyl)和核糖(Nbo)。红外光谱分析表明,PNMPⅢ-a是一种含有氨基、硫酸酯键、β-糖苷键和α-D葡萄糖的蛋白多糖。

Abstract:The extraction, isolation , purification and identification of water-soluble polysaccharide from mycelium of Pleurotus nebrodensis were studied. The results showed that after DEAE-Cellulose 52 and Sephadex G- 200 column chromatography, single component PNMP Ⅲ- a was got. PNMP Ⅲ- a was identified by gelose gelatin elect rophoresis,UV spectrum scanning and Sephadex G-200 column chromatography as homogeneous.The results showed that PNMPⅢ-a was a pure polysaccharide .Through the physicochemical characteristics analysis of the polysaccharide, PNMPⅢ-a was a non-starch pure polysaccharide without disaccharide,glucuronic acid and nucleic acid.Its averagemolecular weight was 15213Da.The results analyzed by HPLC after complete hydrolysis by acid showed that PNMP Ⅲ- a was mainly consisted of glucose, rhamnose, xylose, and ribose. IR analysis revealed thal PNMPⅢ-a was protein-polysaccharide containing amido,sulfated bonds,β-1 ,4 glycosidic bonds and α-D glucose.
