葛根淀粉提取及直链、支链淀粉的双波长测定

摘    要:以淀粉提取率为指标,采用正交实验研究了葛根淀粉的加工工艺,并确定了最佳工艺参数为:料水比为2:17,浸泡液的pH为8.5,浸泡时间为1h;同时采用双波长分光光度法对葛根中直链淀粉和支链淀粉的显色反应和光度测定的最佳条件进行了详细的考察,根据吸收光谱,确定直链淀粉的测定波长为617nm和476nm,支链淀粉的测定波长为415nm和532nm,建立回归方程并求得葛根淀粉中直链淀粉的含量为18.29%,支链淀粉的含量为77.24%.

Abstract:The extraction technology of Pueraria lobate starch was studied in the paper.Starch extracting ratio was used as test index.Orthogonal experiments showed that the optimum technology parameters were ratio of materiel and water of 2: 17,the marinating time of 1 h,the pH of 8.5.Dual-wavelength spectrophotometry was applied to the determination of amylase and amylopectin in Pueraria Iobata.According to the absorption spectra of the two complexes,the wavelength of l_2-amylose,617nm and 476nm, and of l_2- amylopectin,415nm and 532nm, were selected as the measuring wavelength .The content of amylase and amylopectin were further determined as 18.29% and 77.24% through standard curve.
