摘    要:本文选取AlCl3分光光度法和反相高效液相色谱法测定薇菜黄酮的含量,旨在确定两种方法的最佳检测条件参数。通过对两种方法涉及的关键参数优化,确定AlCl3分光光度法的最佳条件:以芦丁为标准品,在357 nm下测定,测得薇菜中黄酮的含量为0.746 mg.g-1;反相高效液相色谱法最佳条件:以甲醇-0.04%磷酸(40∶60/v∶v)为流动相,等度洗脱,360 nm下测定,测得薇菜中黄酮的含量为0.798 mg.g-1。反相高效液相色谱法测定精度高,重现性好,但仪器和试剂价格昂贵;AlCl3分光光度法虽然专一性稍差,但它不需要价格昂贵的仪器和试剂,一般实验室均可完成。

Abstract:To select the optimum detection condition,this paper chose AlCl3 spectrophotometric and RP-HPLC methods to detect the content of the osmund flavones.The results showed that the optimal conditions of the AlCl3 spectrophotometric method are based on rutin as standard,and detection wavelength is 357nm.Under these conditions,the flavanone content of the osmund is 0.746 mg·g-1;the optimal conditions of RE-HPLC are the methylalcohol-0.04% phosphoric acid(40∶60 /v∶v) for flows,and same density eluted under 360nm.Under these conditions,the flavones content of the osmund is 0.798 mg·g-1.The accurary is very high by RP-HPLC method,and this means has better reproducibility,but RP-HPLC method needs expensive apparatus or regents;The AlCl3 spectrophotometric method has poor specificity,but it does not require expensive equipment and regents,so many laboratories can provide the requirment for the method.
