摘    要:采用HPLC-DAD-APCI-MS对叶黄素及其酯衍生物进行定性和定量分析。通过比较保留时间、紫外可见光谱图和质谱图,鉴定了叶黄素二琥珀酸酯、叶黄素单琥珀酸酯和叶黄素的化学结构。建立了叶黄素二琥珀酸酯的C30-HPLC检测方法,以0.05%乙酸甲醇溶液-0.05%乙酸乙腈溶液(50∶50,V/V)为流动相,在1050μg/mL范围内峰面积和进样量呈良好的线性关系,且具有较高的准确度和良好的重现性。该方法经济可靠,可准确监测合成、加工及贮藏过程中叶黄素二琥珀酸酯的变化。

Abstract:Objectives of this study were to develop a high performance liquid chromatography-photo diode array detection-mass spectrometry (HPLC-DAD-MS) method with atmospheric pressure chemical ionization (APCI) mode for qualitative and quantitative analyses of lutein and its ester derivatives.Lutein disuccinate, lutein monosuccinate and lutein could be identified according to the combined use of retention times, UV-visible absorption spectra and mass spectra.An accurate and reproducible C30-HPLC method of lutein disuccinate was achieved with a binary mobile phase consisting of 0.05% acetic acid in methanol -0.05% acetic acid in acetonitrile (50/50, v/v) in the linear range of 1050μg/mL.This economical and effective method could be used to monitor its change during synthesizing, processing and storage.
