摘    要:目的提高肾上腺素SOD测活性法的灵敏度。方法选用新的测定波长和反应测定体系,提高测定的灵敏度。同时研究了肾上腺素浓度、pH、温度及金属离子对自氧化速率的影响。结果于波长325nm处检测的灵敏度高于改进前在480nm处检测的灵敏度。增加底物浓度、提高pH及升高温度都能使自氧化速率增加。用改进后的方法对SOD标准品进行测活性,平均回收率为93.32%,批内变异系数(CV)7.71%,最大抑制率为70.30%。结论改进后的测定系统具有更高的测定灵敏度,且重复性好、准确度高。

Abstract:Objective To increase the sensitivity of the epinephrine autoxidation method for the determination of superoxide dismutase(SOD) activity.Methods The new wavelength and reaction system for the SOD activity determination were adopted.The influential factors including concentration of epinephrine,pH,temperature andmetal ion were studied for their influnce on the autoxidation rate.Results The sensitivity determined under thewavelength of 325nm was higher than that under 480nm.With the increase of substrate concentration,pH andtemperature,the autoxidation rate rose accordingly.Measuring the SOD standard preparation with this improvedmethod,the average recovery rate and coefficient of variation(CV) in batches are 93.32% and 7.71%,respectively,and maximal inhibition rate is 70.30%.Conclusion The improved determination system is more sensitive,repro-ducible and exact.
