摘    要:本文对用GC-MS法测定血浆中~(15)N-甘氨酸进行了初步探讨。实验主要内容包括:(1)两种氨基酸衍生物(N-三氟乙酰甘氨酸正丁酯和N-七氟丁酰甘氨酸异丁酯)的制备;(2)混合氨基酸衍生物的分离;(3)同位素丰度的GC-MS测定。方法较为简便,灵敏、省时和精确。只需0.5ml全血即可满足一次样品的测定要求。本法对单纯甘氨酸样品测量的精确度为0.6%,混合氨基酸样品测定的实际误差在3%左右。

Abstract:A method for the determination of 15N-glycine in plasma by GC-MS is dealt with. The procedure consists of: (1) Preparation of N-heptaf luorobutyl-glycine isobutyl ester and N-trifluoroacetyl-glycine n-butyl-ester; (2)Separation of mixed amino acid: (3)Detetrm-ination of isotopic abundance by GC-MS. This method is simple sensitve, accurate and timesaving.0.5ml of whole blood is sufficient for each determination.The accracy is 0.6%for glycine and 3% for mixed amino acid.
